We reported in @ACSSynBio part of my masters work on CRISPR-based diagnostics. We coupled three thermostable Cas effectors to LAMP to detect three targets in a single step reaction. We assumed AapCas12b cuts DNA, but we proved it wrong. doi.org/10.1021/acssyn… 1/4

We found out that AapCas12b cuts RNA by collateral cleavage, making it interfere with TccCas13a. So we purified BrCas12b, another thermostable Cas effector. However, it also cuts some RNA reporters. This collateral cleavage of RNA may be universal to the Cas12b family. 2/4
