It’s my absolute pleasure and utmost honor to share our recent publication in the @BloodPortfolio of the @ASH_hematology , titled:
“Molecular Consequences and Targeting of PPM1D Overexpression in Myeloproliferative Neoplasms,” with me as
@DarioAmodei We are focused on BioMedical AI development and we agree with Dario. We need to be careful , evaluate at full extent and perform measured RSI. Thank Dario. Ready to work with you.

Excited to share our new publication at @CommsBio ( @Nature ), “Resistance to modulation of in vivo gene expression by genomic editing.”
The study took long time and enormous effort of a whole team of investigators!
We investigated whether CRISPR editing of KLF1 regulatory enhancers (EHS1 and INT1) could reproduce KLF1 haploinsufficiency and increase fetal hemoglobin (HbF/γ-globin)!
1. Although enhancer editing reduced KLF1 and increased γ-globin in primary human CD34+ cells, these effects were not consistently maintained following xenotransplantation—revealing an unexpected resilience of the KLF1 regulatory locus in vivo.
2. Our ATAC-seq studies suggest that dense regulatory architecture, enhancer redundancy, and long-range chromatin interactions can buffer gene expression even after efficient genome editing. This highlights an important principle for therapeutic genome engineering: editing an enhancer does not necessarily phenocopy natural genetic haploinsufficiency.
Grateful to our collaborators and especially pleased to have contributed as a co–equal contributor to this work. These findings may help guide the design of more durable and predictable genome-editing strategies for hemoglobinopathies and other genetic diseases. #GenomeEditing#CRISPR#GeneTherapy#KLF1#Hemoglobinopathies#FetalHemoglobin#Hematology#StemCells#Epigenetics#ATACseq#CommunicationsBiology
