How does optical resolution affect the spatial precision of two-photon optogenetic photostimulation? 👇Read on & see our preprint to learn more! biorxiv.org/content/10.110…
@paqio Thanks for doing these very useful tests for holographic optogenetics! I was wondering whether you also considered the PSF broadening due to scattering in the brain. Do you think that scattering could also be a factor that can dominate PSF extent (instead of the microscope PSF)?
@ptrrupprecht It should be a factor, especially when targeting deeper structures, though we don't have the data to address this directly. In this study, we compared two different PSF sizes, 8 and 19 um (axial FWHM). Do you think the broadening would affect these two sizes very differently?
@paqio @ptrrupprecht @FavrebulleItia has done work on this question if you want to check it out
@DrYassum @paqio @FavrebulleItia Thanks for pointing this out (nature.com/articles/srep1… ?). It seems interesting but leaves also several questions open, e.g., how this translates to much deeper imaging in mouse cortex, using 2P instead of 1P holographic patterning.
