Another novelty in Bambu to highlight is that it could accurately quantify transcripts after transcript discovery, with both full-length and unique read counts provided, shown to be more sensitive filters for unexpressed transcripts than conventionally used estimates. 3/4
The main novelty in Bambu is that it uses a machine learning model to predict transcript probability score, later calibrated into a single interpretable, scalable, Novel Discovery Threshold (NDR), shown to outperform conventional thresholds (readCount, GeneReadProp, etc). 2/4
