Healthspan Edge @NotDead_X

Growth signaling cut, female mice lived 8 percent longer. Mouse study, out this September in Aging Cell from John Kopchick's lab at Ohio University. The receptor for growth hormone was switched off at 12 months, roughly middle age, in 30 to 35 animals per sex per group. In females, median lifespan rose 8 percent, from 929 to 1,003 days. The p value was 0.049, just under the usual 0.05 cutoff for calling a result significant. Males gained 7 percent, which did not reach significance on that measure. The paper's title says it extends healthy lifespan. In females, the healthy part did not show up: no gains in grip strength, balance, frailty or bone. Males improved on insulin sensitivity, grip and balance, and both sexes carried more body fat. Anti-aging clinics have sold growth hormone injections for years, pushing that signal up. These mice lived longer with it turned down. In the US, distributing growth hormone for anti-aging is a federal felony. The classic version, mice born without the receptor, has been one of the most replicated lifespan results in mammals since the late 1990s. Switching the receptor off in midlife, the only timing a human drug could realistically copy, gives a much smaller effect. Humans with the same receptor defect exist. In Ecuador, about 100 people born with it (Laron syndrome) were followed for 22 years, and none developed diabetes, against 5 percent of their relatives. Cancer appeared once, and it was not fatal, against cancer in 17 percent of relatives. They did not live longer, mostly because of accidents, alcohol and other causes unrelated to age. This is observational data, not a trial. So the pathway with nearly three decades of mouse data, switched off in midlife, produces a modest, borderline result. Meanwhile the field's hottest idea, cell reprogramming, got New Limit $435 million in June at a $3.1 billion valuation, with no human data yet and a first trial planned for 2027. A drug that turns growth hormone signaling down in midlife without the extra fat would be a real business. That is a concept. Nobody has one built for aging yet. A modest effect in one cohort of mice, with a cost attached. An 8 percent gain in one sex at p equal to 0.049 is a reason to repeat the experiment, and not a reason to buy anything a clinic is selling.

@bensmithlive The same research credits antibiotic eradication with about 36 percent fewer stomach cancers.

@codewhatyouwant half right. females weren't healthier, males were, on insulin and grip

@BrandonLuuMD More ink did not mean more risk. No dose response. That is the puzzle.

@fmfclips Key word is "associated." Observational data, not a trial. Still a useful exchange rate.

@realDailyWire Marital conflict shows up in blood work, not just at dinner. Kiecolt-Glaser, 2005.

2.61 is from session four. The sales page says six.

Plasma swaps cut 2.61 years of biological age, briefly. That's a 42-person pilot in adults over 50, published in Aging Cell and measured on aging clocks in the blood rather than on disease or lifespan. And the company selling the procedure advertises the result at exactly the point where it disappeared. The procedure is therapeutic plasma exchange: blood is drawn and its plasma swapped out for a 5% albumin solution before the blood goes back in. Circulate Health funded the study, and three of the authors are Circulate members. Circulate's site advertises "an average 2.6-year reduction in biological age after just six TPE sessions." Six sessions is where it breaks. The 2.61 comes from the measurement after session four, in the group that also got IVIG, pooled donor antibodies given by infusion. After session six, the preprint of the same trial reports no significant difference from a sham procedure in any group. The published paper leads with the earlier timepoint, and the authors suggest the body compensates. In August Viome bought Circulate, and the acquisition release repeats the 2.6. Sessions have run about $8,000 to $10,000 each by GeekWire's reporting, so a six-session course comes to roughly $48,000 to $60,000. Next Health sells its own six-session bundle for $45,000. My objection is narrow. Plasma exchange is an established procedure for specific diseases, and this single-blind trial logged one mild allergic reaction across 240 procedures. The Mayo Clinic clip in the first comment shows that established use, with neurologist Brian Weinshenker on multiple sclerosis and neuromyelitis optica: 1:13 the clinical background 3:04 the plasma exchange itself The objection is to the number: a lab clock instead of health outcomes, about ten people in each of four groups, a headline figure that needed IVIG, and an effect that did not survive to the session count the sales page advertises. What would settle it is a larger sham-controlled trial, run by people who do not sell the sessions, that still shows the drop after session six. Until somebody runs it, 2.61 is a four-session number.

Dead bacteria made old mice look younger on paper. Nature Aging published the study on September 24, 2026. It comes from Jing-Dong Han's lab at Peking University, and the organism is Neisseria flavescens, a bacterium that already lives in your mouth. No human picked this bacterium. The team built an AI aging clock they call AURORA, had it model what would happen if you added or removed each microbe, and this one came out on top as the predicted winner. Screening by model is normal practice and fine on its own. What matters is what the screen got followed with. In people, nothing. The human data is associative, with no intervention at all. The closest thing to a longitudinal check tracked 132 participants over a year, and it is still a correlation. And in the study's own supplementary figures, when they ranked people directly by how old they looked biologically, N. flavescens did not come out as a top correlate. The headline exists because the model put it there. In worms, live bacteria did extend lifespan and healthspan. Movement was scored on 30 worms per group. In mice it gets thin. Heat-killed bacteria, ten weeks, 20-month-old animals, six per group and five in some panels. Blood chemistry and liver gene activity shifted toward the patterns of younger mice, and the gut bacteria shifted too. Survival was never measured, and there is no mouse lifespan curve anywhere in the study. Food intake was recorded per cage with no statistics run on it. The supplementary notes say the P values weren't corrected for testing many things at once, which is how findings that aren't there get published. The authors' own figure title calls the reversal partial. Worms got live bacteria and mice got dead ones, and the sections I can read don't say why the switch was made. That's the first thing I'd want answered. No company is attached to this, and no investor. The funding is Chinese public money. The one commercial fact in the study is a pending patent, number 202611084872X, filed by the senior author and a colleague and not yet publicly viewable. It was filed before a single human had been given anything. The selling started years ago regardless. Viome runs a $399 test that reads your oral microbiome and hands back a biological age score, plus lozenges at $79 a month on subscription. Pendulum sells Akkermansia muciniphila for $54, and Akkermansia turns up inside this very study as a loose correlation rather than a result. Bristle sells a $169 oral microbiome test and makes no aging claim at all, which deserves credit. Keep the two markets apart. The longevity economy means everything sold to older people, and it is enormous. Biotech that actually treats aging is a much smaller thing that mostly hasn't worked yet. The products above sit in the first market while borrowing the language of the second. There's a history here too. In 2018 this same bacterium was pulled from gastritis patients and written up in Current Microbiology as a possible pathogen, with the authors asking someone to study whether it causes disease. Nobody appears to have run that study, and nobody has established what a live strain does in a person. This is worth watching and not worth buying. An AI model nominated the bacterium, worms lived longer on it, and six old mice per group had younger-looking blood work after ten weeks of dead cells. That's a hypothesis with a patent attached, and the readout that matters is whether anyone runs it in living humans. Zero survival curves in the study. One $399 test already on the shelf.

Patent filed before a single human received anything.

XPRIZE paid $1 million. Did anyone check the trial? On August 11 the $101 million XPRIZE Healthspan named ten finalist teams and paid each of them $1 million. One of the ten is Longeveron, in on the strength of a stem cell trial in aging frailty. The ceremony recording opens on an empty room. 6:28 is where it actually starts. The trial is real. Phase 2b, 143 people in the final analysis, ages 70 to 85, peer reviewed in Cell Stem Cell in February. One infusion of donor mesenchymal cells, and at nine months the treated group walked 63 meters further in a six minute walk test than placebo, p equals 0.0077. Now open the registry. NCT03169231 states the primary endpoint plainly: change in six minute walk distance at 180 days. At 180 days the result was p equals 0.0635. In trial language, that is a result you are not allowed to claim. The number everyone quotes comes from a later time point than the one the trial registered. That is not fraud and it is not nothing. It is a trial that missed its primary endpoint and found significance three months past it, and that distinction is the whole difference between a result and a lead worth chasing. Two things the paper says about itself. From the limitations: the TNF-alpha measurements, the inflammation marker the authors use to explain how the therapy works at all, were discarded because the samples degraded in storage. From the disclosures: the University of Miami holds equity in Longeveron. The company licenses the university's intellectual property. The senior author is the company's chief scientific officer. The money is where it gets strange. Longeveron closed at an $8.67 million market capitalization on September 22. That $1 million milestone is roughly an eighth of the entire company. And on September 16 a separate Phase 2b of the same cell product, this one in a pediatric heart condition, missed its primary endpoint outright at p equals 0.8336. The company has said its cash runs into the fourth quarter. What the trial did not show is longer than what it did. A walking test measures distance. It does not measure years. People with dementia were excluded, so cognition is untouched. And nobody has explained why a single transient infusion would do more at nine months than at six. The business here is not a drug. XPRIZE had to build a standard set of healthspan measurements because none existed: muscle function, cognition, immune markers, with the thresholds for what counts as meaningful still being written. Until a regulator recognizes aging as an indication, the dependable money in this field is in selling the measurement, not the molecule. There is a smaller job hiding in the same story: reading the registry entry before the press release, and noticing when the time frame moved. The trial that would settle this starts enrolling in summer 2027 and hands data to the judges in 2029. Until then, 63 meters at nine months is a reason to run it, not a reason to believe the answer.

@dontbelievetoAI The skipped chapter is a registered endpoint and years of follow-up, not weeks

Diet study sold as age reversal never measured aging. The Sydney trial spread as four weeks of eating turning back biological age. It is a genuine randomized controlled trial, published in Aging Cell in April 2026, open access. Humans, not mice. 104 adults aged 65 to 75, four weeks, four diet arms. Two were higher in fat and two higher in carbohydrate, and each pair was also split between an omnivorous and a vegetarian protein source. What it measured is the part that fell out of the coverage. Not an epigenetic clock. It used the Klemera-Doubal method, a score built from a routine blood and clinical panel: cholesterol, insulin, CRP and the rest of what an annual physical already measures. A legitimate research tool, and a different instrument from the DNA methylation clocks that the phrase biological age puts in people's heads. The result, in the paper's own terms. The arm closest to how these people normally ate showed no significant change in that score. One of the higher-carbohydrate arms scored significantly lower than that reference arm. The remaining two moved in the same direction, and not all of those shifts reached statistical significance. Then the authors do what the coverage skipped. They warn against their own headline. Caution is warranted, they write, in reading such changes as evidence of biological age reversal, because the shifts may reflect acute physiological responsiveness to dietary inputs rather than altered ageing trajectories. Translated: your blood panel moved. That is a smaller claim than your ageing slowed. The registry is where this gets sharp. The parent study was registered in 2016 with 129 people at baseline, and its declared primary outcomes are appetite, cardiometabolic health, oral health, microbiota and body composition. Biological age is not among them. Declaring your outcome before you collect the data is what stops a study from fishing, and this ageing question was attached to the dataset roughly ten years after it was gathered, in a re-analysis of 104 of those people. Protein was also held at 14% of energy in all four arms, which makes the trial structurally unable to say anything about how much protein anyone eats. I want to be precise about my own objection. It is not that they used the wrong clock. Eric Verdin of the Buck Institute has said his own age comes back anywhere from 40 to 67 across methylation tests, so the clock people imagine is no steadier than the one actually used. The problem is that nobody ran a study designed to answer this question, and a four-week readout got reported as though somebody had. The money sits in a third market that keeps getting folded into the other two. Not the longevity economy, which counts everything people over 50 spend and runs into the tens of trillions. Not the biotech trying to treat ageing, which runs in the tens of billions. Biological-age testing is its own small market. One research firm put it near $0.58 billion for 2026 and about $0.99 billion by 2030, in a release this September. Two weeks earlier a different firm projected $5.58 billion for that same year, 2030. Two vendors selling forecasts on one market, five and a half times apart. Retail is firmer than either forecast. A TruAge Complete panel runs $499, per Forbes in February 2026, which is roughly what this entire market rests on: people paying real money for an estimate with no validated outcome behind it. What would settle any of this is a trial that registers biological age as its primary outcome, in a clock validated against real health outcomes, running for years instead of a month. Nobody has run it. Until somebody does, every number in this market is a proxy for a proxy, and that includes the reassuring ones.

abstract is worth the two minutes on its own. The authors' caution paragraph is the part almost nobody quoted

Aging cells just stole a trick from cancer. Cedars-Sinai researchers found that senescent cells, the worn-out "zombie" cells that pile up as we age, make a protein called PD-L2. Tumors use that exact same protein to tell the immune system to back off. Block PD-L2 in old mice and the zombie cells stop piling up so fast. The mice processed sugar better and gripped objects with more strength, both real, measurable markers of aging in a mouse. Human tissue told a similar story. Aged cells made more PD-L2 than young ones. That's a comparison, not a treatment. Nobody has blocked this protein in a living person. Not yet. Eighteen separate NIH grants paid for this one paper. On top of that sits money from Hevolution Foundation, the Saudi-backed group that has said it wants to spend a billion dollars a year chasing exactly this kind of biology. That's public and philanthropic science money funding a mouse study. It's not the same pot as the $27 trillion longevity economy. That number is what older people already spend on healthcare and everything else, not biotech research. Two different numbers, two different games. Eighteen grants and a Saudi foundation's money is a reasonable bet on curiosity. It is not evidence that blocking this protein does anything in a person. Cell Metabolism published this on September 10. Right now it lives in mice and lab dishes, nowhere else.

Old mice on Ozempic just outlived the hungry ones. Twenty-month-old female mice on semaglutide lived a median 834 days. Salt-water controls got 742. That's a mouse study, from UC Berkeley and the National Institute on Aging, published in Nature this month. Semaglutide also out-performed a strict 24 percent calorie cut on memory and blood sugar, though the starved mice matched it on most other markers, and every mouse in the group was female. Different paper, different species. 108 adults on semaglutide for a fat disorder linked to HIV slowed their DunedinPACE clock by 9 percent, in a randomized trial. TruDiagnostic, the company that sells that clock, co-authored the study. Its own lead researcher called it a signal, not a reversal. This drug class pulled in $79 billion in 2025 combined, all of it for weight loss and diabetes. Add aging as indication three and nobody needs a new molecule to sell it again.

$700 million biotech built on mice just died. Unity Biotechnology co-founder Jan van Deursen ran the mouse study himself. He and his Mayo Clinic team gave old mice a genetic switch that killed off p16-positive senescent cells. Median lifespan stretched by as much as 35 percent. Nature published it in February 2016, seven years after he helped start the company betting on it. By October 2016, Bezos, Thiel, and Mayo Clinic itself had put $116 million behind that bet. Unity IPO'd in 2018 at a $700 million valuation on $85 million raised. Its lead drug, UBX0101, took the same trick into a human knee. One injection, aimed at killing senescent cells to ease osteoarthritis pain. Phase 2, not approval, read out in August 2020. 183 patients, zero difference from placebo. Stock dropped 60 percent that morning. Nine years after the mouse paper, the company is gone. It never found a human disease where the mouse math held. Layoffs hit the entire staff this May. Liquidation finished by September 2025. The mouse result still holds up. Nobody has repeated it in a human knee, or anywhere else in a human body. That gap, between a cured mouse and a paying patient, is where most of this industry's money sits.
